How do I know how far out ot place drain line on new construction for toilet? Also if the drain is tied in to an existing bathroom line do I have to run a new vent?

12" from the wall is a standard rough in dimension. jusy make sure you dont buy a 14" rough in toilet. As far as venting goes, you will probably have to add a new vent. I am assuming you are not adding a toilet right next to an existing one, or on the other side of the wall. You will need to use atleast a 2" vent line. Tie it into the main stack in the attic so you dont have to make another roof penetration.
